Drug Release from Hollow Type Suppository(¥±)


Abstract
In order to evaluate an Indomethacin(IDM) Polyvinyl alcohol(PVA) Hydrogel as a sustained-release vehicle for rectal administration, the in vitro release characteristics of IDM from hydrogel were examined. The 5 types of hollow type suppositories and one type of conventional suppository were prepared with PVA hydrogel by a low-temperature crystallization method.
Slower release of IDM occurred at the higher PVA concentration and the amount of the cross-linking agent of a glutaraldehyde(GA). The drug amount released(£¥) from type V, hollow type suppository containing 12.5§· of IDM powder dispersed in the base and 0.5§¢ of 6.25£¥(w/v) IDM-PEG 300 solution in the cavity, was high as about 95£¥ and the release rates were more rapid at early time than any other suppositories.
For comparative study, IDM suppository was prepared with Witepsol H-15 using the fusion method. Amounts of IDM released from hydrogel showed a sustained released compared with Witepsol H-15 suppository.
