Drug Release from Hollow Type Suppository(Ⅱ)

Abstract
|
|
|
In order to evaluate an Indomethacin(IDM) Polyvinyl alcohol(PVA) Hydrogel as a sustained-release vehicle for rectal administration, the in vitro release characteristics of IDM from hydrogel were examined. The 5 types of hollow type suppositories and one type of conventional suppository were prepared with PVA hydrogel by a low-temperature crystallization method.
Slower release of IDM occurred at the higher PVA concentration and the amount of the cross-linking agent of a glutaraldehyde(GA). The drug amount released(％) from type V, hollow type suppository containing 12.5㎎ of IDM powder dispersed in the base and 0.5㎖ of 6.25％(w/v) IDM-PEG 300 solution in the cavity, was high as about 95％ and the release rates were more rapid at early time than any other suppositories.
For comparative study, IDM suppository was prepared with Witepsol H-15 using the fusion method. Amounts of IDM released from hydrogel showed a sustained released compared with Witepsol H-15 suppository.
